This project uses modern biochemical methods to isolate suprastructural elements and authentic aggregates of structural proteins from the papillary dermis, the dermal-epidermal junction or the cytoskeleton of basal keratinocytes. In collaboration with other network partners, suprastructures from laminin a3-deficient skin and keratinocytes, from the skin of mice with keratin mutations and of the hypomorphic dystrophic EB mouse will be isolated and characterized. The same techniques will be used after therapy of the mouse models with molecular chaperones or cell based therapies, in order to identify restorative effects at the suprastructural level.
